When I open the LightGen window, no matter what settings I choose, the Generate Variants button remains disabled. I’ve not activated LightGen previously in this model but have done it successfully in other models in the past. Am I doing something wrong or missing a step? I don’t do any specific prep before opening LightGen.

Are you trying to Generate Light Gen scenarios while Interactive render is enabled perhaps? If yes, please note that Light Gen cannot be generated during render.
You can also hover your mouse over the button to see a message that appears.
