How to manually register the Win service after installing OLS?

Is it only necessary to register from the installer?

For example, after Win10 update, I want to re-register from command instead of reinstallation.

I'd recommend you to install the license server anew to be sure that it'll work correctly. If you have the latest license server (4.5.1) you won't experience the problem with Windows updates.
If you want to install it command-line (no GUI) you can use -gui=0 argument. It will generate a config file with the options chosen that you can use for a silent install - you can check the example file attached.
For example - a command line install would look like (use full file paths if the installer .exe and config.xml are not both in the folder you've navigated to in CMD):

vrlservice_win_4.5.1.exe -gui=0 -configfile=config.xml

I was relieved to find out that the latest license server is taking measures against Windows update.
