In the render globals, under the DoF settings, is the Aperture Size parameter used to calculate the diameter of the aperture or is it the radius?

Aperture (in the Render Globals roll-out) is the size of the virtual camera aperture in world units, meaning that it has no real-world counterpart. Small aperture sizes reduce the DOF effect, larger sizes produce more blur.
Instead of using Render Globals roll-out, you could enable DOF in the V-Ray Physical Camera parameters and use F-number parameter to control the aperture size of the virtual camera. F-number represents the ratio of the diameter of the lens aperture to the length of the lens. Lowering the f-number value increases the aperture size therefore the Depth of Filed, in reverse, increasing the f-number will reduce the DOF effect.
Also, keep in mind that when the DOF in V-Ray Physical camera is enabled it will override the Depth of Field in the Render Settings.
